LABORATORY WORK

1. Quantification of total lipids in the blood

The products of unsaturated lipid decomposition interact with phospho-vanillin reagent in the presence of sulfuric acid to yield a product with characteristic pink color. The color intensity is proportional to the lipid content in the analyzed sample.

Procedure

► Prepare the control, experimental and standard samples.

► Put 0.1 ml of blood serum into the experimental sample (tube 1), put 0.1 ml of distilled water into the control sample (tube 2), and put 0.1 ml of standard triolein solution into the standard sample (tube 3).

► Add 2.9 ml of concentrated sulfuric acid to all three test tubes.

► Mix the contents of all tubes intensively and place them in boiling water for 10 min.

► Remove the test tubes from the water bath and cool under running tap water.

► Put 0.2 ml of aliquot from each test tubes into three new test tubes containing 3 ml of phosphovanillin reagent.

► Mix the samples intensively by shaking and then put in a dark place.
